Kayu kuning Arcangelisia flava (L.) Merr merupakan salah satu tanaman yang dapat dijadikan obat tradisional untuk mengobati berbagai penyakit. Selain alkaloid, kayu kuning juga diketahui mengandung senyawa flavonoid yang berguna bagi kesehatan. Penelitian ini bertujuan untuk mengidentifikasi kandungan senyawa flavonoid pada ekstrak etanol batang kayu kuning Arcangelisia flava (L.) Merr dengan metode kromatografi lapis tipis. Sampel awalnya dimaserasi dengan pelarut etanol selama 3x24 jam, kemudian filtrat yang diperoleh diuji pendahuluan flavonoid dengan menambahkan beberapa ml etanol dan beberapa ml HCl pekat. Hasil uji pendahuluan, menunjukan sampel positif mengandung flavonoid ditandai dengan perubahan warna larutan menjadi merah tua. Setelah itu dilakukan uji penegasan senyawa dengan metode kromatografi lapis tipis. Hasil uji KLT menunjukan bahwa ekstrak etanol batang kayu kuning positif mengandung senyawa flavonoid dengan fase diam silica gel 60 F254 dan fase gerak metanol:kloroform:etil-asetat (2:1:1) terjadi pemisahan yang baik dan menghasilkan bercak berfluoresensi dibawah UV 366 nm dengan nilai Rf 0,60.

Yellow wood (Arcangelisia flava (L.) Merr is a plant that can be used as a traditional medicine to treat various diseases. Besides alkaloids, yellow wood is also known to contain flavonoid compounds that are useful for health. This study aims to identify the content of flavonoids in ethanol extract yellow sticks Arcangelisia flava (L.) Merr by thin layer chromatography method. Samples were initially macerated with ethanol for 3x24 hours, then filtrate obtained preliminary flavonoids tested by adding a few ml of ethanol and a few ml of concentrated HCl. Preliminary test results, the samples showed positive for flavonoids characterized by discoloration of the solution became dark red. After confirmation of the test compounds was performed by thin-layer chromatography method. The test results showed that the ethanol extract TLC dowel positive yolk contains flavonoids with the stationary phase of silica gel 60 F254 and a mobile phase of methanol: chloroform: ethyl acetate (2: 1: 1) there is a separation of good and fluoresces under UV spotting produce 366 nm with the Rf value of 0.60.
