Abstract
Microspores have the potential to develop into new individuals when cultured in an appropriate medium, as each of these cells possesses totipotency. This study aims to analyze the developmental stages of microspores in the flower buds of kaffir lime (Citrus hystrix) plants of varying sizes. This stage is a crucial initial step before initiating a plant breeding program using anther or microspore culture techniques. In this study, the developmental stages of microspores were observed in 13 flower buds measured for length. Microspores were examined using a simple method: two anthers were crushed in a petri dish containing five drops of sterile water, and the mixture was then transferred to a glass slide for microscopic observation. Additionally, morphological observations of the anthers were conducted, along with counts of the number of anthers and petals in each flower bud. The results showed that among the 13 flower buds observed, not all stages of microspore development were found. The mature pollen stage was more frequently observed. These findings can serve as a reference to optimize the identification process of kaffir lime flower bud morphology that contains microspores as an initial step for initiating microspore culture.
